演題：Identifying the cancer mechanisms and novel drug candidates through a whole-body phenotypic screening

Abstract：
Animal models have revealed critical cancer vulnerabilities. To accelerate drug discovery, we developed multigenic Drosophila strains modeling patient-specific genetic abnormalities, integrated with mammalian systems. Leveraging this platform, we established a novel method to reduce drug toxicity through determining an “anti-target” of a kinase inhibitor causing toxicity, enabling rational drug redesign. Additionally, to tackle pancreatic ductal adenocarcinoma (PDAC), we created models mimicking diverse genetic alterations. Genetic screening using these models pinpointed key kinases driving transformation, which were effectively targeted by inhibitors to suppress human PDAC xenografts. These platforms provide efficient, complementary tools for elucidating cancer mechanisms and identifying therapeutic strategies.
